What is this medicine used for?

This medicine is taken by mouth to treat certain kinds of myelofibrosis (a disorder of the bone marrow).

This medicine may also be used to treat other conditions as determined by your healthcare provider.

What should my healthcare provider know before I take this medicine?

Before taking this medicine, tell your healthcare provider if you have ever had:

• an allergic reaction to any medicine, to benzyl alcohol, or to yeast
• liver or kidney disease, or if you are on dialysis

Also, tell your healthcare provider if you have recently had any kind of infection, including fungal or viral infections.

Females of childbearing age: Talk with your healthcare provider if you are pregnant or plan to become pregnant. It is not known whether this medicine will harm an unborn baby. Do not breast-feed while taking this medicine without your healthcare provider's approval.

How do I use it?

Check the label on the medicine for directions about your specific dose. Take this medicine exactly as your healthcare provider prescribes. Your healthcare provider may adjust your dose based on your lab tests. Do not stop taking this medicine without your healthcare provider's approval. Do not miss doses or take more than prescribed.

You may take this medicine with or without food. Taking it with meals may lessen the chance the drug will upset your stomach.

What if I miss a dose?

If you miss a dose, skip the missed dose and take the next one as directed. Do not take double doses. If you are not sure of what to do if you miss a dose, or if you miss more than one dose, contact your healthcare provider.

What if I overdose?

If you or anyone else has intentionally taken too much of this medicine, call 911 or go to the emergency room right away. If you pass out, have seizures, weakness or confusion, or have trouble breathing, call 911. If you think that you or anyone else may have taken too much of this medicine, call the poison control center. Do this even if there are no signs of discomfort or poisoning. Symptoms of an acute overdose may include: shortness of breath, nausea, fever, rash, fast heartbeat, headache, chills.

What should I watch out for?

You will need to have blood tests regularly to see how this medicine affects you. Keep all your appointments for treatments and tests.

You may get infections more easily when you are taking this medicine. Stay away from people with colds, flu, or other infections. Also, do not have any vaccines without getting your healthcare provider's approval first.

If you need emergency care, surgery, or dental work, tell the healthcare provider or dentist you have received this medicine.

What are the possible side effects?

Along with its needed effects, your medicine may cause some unwanted side effects. Some side effects may be very serious. Some side effects may go away as your body adjusts to the medicine. Tell your healthcare provider if you have any side effects that continue or get worse.

Life-threatening (Report these to your healthcare provider right away. If you cannot reach your healthcare provider right away, get emergency medical care or call 911 for help): Allergic reaction (hives; itching; rash; trouble breathing; tightness in your chest; swelling of your lips, tongue, and throat).

Serious (report these to your healthcare provider right away): Fever; chills; sore throat; unusual weakness or tiredness; shortness of breath; cough; unusual bruising or bleeding; painful skin rash or blisters; severe nausea or vomiting; pain or burning with urinating.

Other: Dizziness, weakness, gas, headache, weight gain.

What products might interact with this medicine?

When you take this medicine with other medicines, it can change the way this or any of the other medicines work. Nonprescription medicines, vitamins, natural remedies, and certain foods may also interact. Using these products together might cause harmful side effects. Talk to your healthcare provider if you are taking:

• antibiotics such as clarithromycin (Biaxin), erythromycin (Ery-Tabs, E.E.S.), isoniazid, metronidazole (Flagyl), norfloxacin (Noroxin), rifabutin (Mycobutin), rifampin (Rifadin), and telithromycin (Ketek)
• antifungal medicines such as clotrimazole, fluconazole (Diflucan), itraconazole (Sporanox), ketoconazole (Nizoral), miconazole (Monistat, Micatin), posaconazole (Noxafil), and voriconazole (VFEND)
• anti-HIV medicines such as atazanavir (Reyataz), delavirdine (Rescriptor), efavirenz (Sustiva), fosamprenavir (Lexiva), indinavir (Crixivan), lopinavir/ritonavir (Kaletra), nelfinavir (Viracept), nevirapine (Viramune), ritonavir (Norvir), and saquinavir (Invirase)
• antiseizure medicines such as carbamazepine (Tegretol), phenobarbital, phenytoin (Dilantin), and primidone (Mysoline)
• antiviral medicines such as boceprevir (Victrelis) and telaprevir (Incivek)
• bosentan (Tracleer)
• conivaptan (Vaprisol)
• heart medicines such as amiodarone (Cordarone, Pacerone), diltiazem (Cardizem, Dilacor, Tiazac), nicardipine (Cardene, Cardene SR), quinidine, and verapamil (Calan, Covera, Isoptin)
• imatinib (Gleevec)
• nefazodone
• St. John's wort
